    为了探究普洱茶茶褐素对脂多糖(LPS)诱导的大鼠肝脏损伤的保护作用。将雄性SD大鼠随机分成正常组(Control)、LPS模型组、茶褐素低、中、高剂量组(即给药量分别为0.135、0.405、1.215 g•kg-1 bw),每三天注射一次LPS(1.5 mg•kg-1)建立大鼠肝脏损伤模型,并进行为期21 d的干预实验。期间记录大鼠的生长状况,实验结束后计算大鼠的肝脏指数并检测肝脏组织中谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,通过HE染色观察肝组织病理变化。检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)活性、TC(总胆固醇)、TG(总甘油三酯)、HDL-C(高密度脂蛋白)、LDL-C(低密度脂蛋白)含量。结果显示,与Control组相比,LPS组大鼠肝细胞排列紊乱、伴有大量炎性细胞浸润及少量肝细胞坏死,脾脏脾小结紊乱及红白髓界限模糊,脾脏指数、肝脏指数、血清中ALT活性和TG含量显著升高(P<0.05),而高剂量的普洱茶茶褐素能够显著降低大鼠脾脏和肝脏指数、血清中ALT活性和TG含量(P<0.05),降幅分别为13.69%、41.10%、41.34%和37.50%,且能缓解LPS诱导的大鼠肝脏和脾脏组织病理损伤。结果表明,普洱茶茶褐素对LPS诱导的大鼠肝脏损伤有一定的保护作用,为普洱茶茶褐素的开发利用提供理论依据。

    Abstract:

    This study aimed to assess of hepatoprotective effect of theabrownin (TB) from Pu-erh tea against lipopolysaccharide (LPS)-induced injury in vivo. A rat model of liver injury was established by injecting LPS (1.5 mg•kg−1) every 3 days. The animals were randomly separated and treated with a placebo, low-dose, medium-dose, or high-dose (0.135, 0.405, and 1.215 g•kg−1, respectively) TB for 21 days. The growth status of the rats was recorded throughout the experiment and at the end of the experiment the liver index was calculated, the enzymatic activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measured in the liver, and histopathological changes in liver tissues were determined by hematoxylineosin staining. Additionally, serum ALT, AST and alkaline phosphatase activities, and serum total cholesterol, total triglyceride (TG), high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol contents were measured. Compared with the control group (healthy animals), LPS-administrated rats exhibited disordered arrangement of hepatocytes, accompanied by extensive inflammatory cells infiltration and focal hepatocyte necrosis, as well as disorganized splenic nodules and indistinct red/white pulp boundaries in the spleen. Furthermore, significant elevations were observed in the spleen index, liver index, serum ALT activity, and serum TG content (P<0.05). High-dose TB significantly reduced the levels of these parameters by 13.69%, 41.10%, 41.34%, and 37.50% (P<0.05), respectively, while also alleviating the LPS-induced histopathological injury in liver and spleen tissues. These results demonstrate that TB exerts some protective effects against LPS-induced liver injury in rats, thus providing a new foundations for the development and therapeutic use of TB.
