    为了研究鼠李糖乳杆菌GG上清液(Lactobacillus rhamnosus GG Supernatant, LGGs)对酒精诱导Caco-2细胞屏障损伤的改善作用,通过构建Caco-2细胞屏障损伤模型,检测分析Caco-2细胞屏障完整性、通透性和细菌移位数量,采用qRT-PCR和Western blot分析LGGs对紧密连接蛋白ZO-1、Occludin和Claudin-1表达的影响,检测活性氧(Reactive Oxygen Species, ROS)、超氧化物歧化酶(Superoxide Dismutase, SOD)和丙二醛(Malondialdehyde, MDA)含量,qRTPCR分析肿瘤坏死因子-α(Tumor Necrosis Factor-alpha, TNF-α)、白细胞介素-6(Interleukin-6,IL-6)、单核细胞趋化蛋白-1(Monocyte Chemoattractant Protein-1, MCP-1)和白细胞介素-10(Interleukin-10, IL-10)的表达水平变化。结果表明:LGGs对于酒精所导致的肠上皮细胞屏障损伤和细菌移位具有修复作用,LGGs干预后ZO-1、Occludin和Claudin-1的mRNA表达水平分别提高了42.02%、45.18%和29.84%,蛋白表达水平分别升高了72.52%、38.72%和31.69%,ROS和MDA含量分别下降了24.92%和30.19%,此外,LGGs的处理降低了TNF-α、IL-6、MCP-1的mRNA表达水平,提高了IL-10的mRNA表达水平。综上所述,LGGs通过增强紧密连接蛋白表达,改善氧化应激损伤和炎症反应来修复肠道屏障损伤,为研发防治酒精性肝损伤的功能性产品提供理论依据。

    Abstract:

    To investigate whether Lactobacillus rhamnosus GG supernatant (LGGs) improved the alcohol-induced barrier damage in Caco-2 cells, Caco-2 cells were used to establish intestinal epithelial cell barrier damage model and detect the integrity and permeability of the intestinal epithelial barrier and bacterial translocation. The effects of LGGs on the expression of tight junction proteins ZO-1, Occludin, and Claudin-1 were analyzed using quantitative reverse transcription polymerase chain reaction (qRTPCR) and western blot. The contents of reactive oxygen species (ROS), superoxide dismutase (SOD), and malondialdehyde (MDA) were detected. The changes of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), monocyte chemoattractant protein-1 (MCP-1), and interleukin-10 (IL-10) were analyzed using qRT-PCR. The results showed that LGGs can prevent alcohol-induced intestinal epithelial cell barrier damage and bacterial translocation. The ZO-1, Occludin, and Claudin-1 mRNA expression levels increased by 42.02%, 45.18%, and 29.84%, respectively, after LGGs intervention. The ZO-1, Occludin, and Claudin-1 protein expression levels increased by 72.52%, 38.72%, and 31.69% respectively. The ROS and MDA contents decreased by 24.92% and 30.19%, respectively. Moreover, the TNF-α, IL-6, and MCP-1 mRNA expression levels were reduced, whereas IL-10 mRNA expression level was elevated. In conclusion, LGGs can attenuate the intestinal barrier damage by enhancing the expression of tight junction proteins, improving oxidative stress damage, and alleviating the inflammatory response. This can provide a theoretical basis for developing functional products against alcoholic liver injury.
