    母乳中的骨桥蛋白(Osteopontin, OPN)与2’-岩藻糖基乳糖(2’-Fucosyllactose, 2’-FL)是调节婴儿肠道免疫及屏障功能的关键活性成分。为了探究OPN与2’-FL组合物对肠屏障功能的改善作用及协同机制,该研究构建了豆粕饮食诱导的斑马鱼幼鱼肠炎模型,用OPN和2’-FL单体及不同比例(1:18~1:1,m/m)组合物进行干预,并测定了肠道中性粒细胞和杯状细胞数量、炎症和肠屏障基因表达水平及细胞因子蛋白表达水平。结果显示,OPN与2’-FL组合物比例为1:9~1:3(m/m)时,能显著降低中性粒细胞数量至每条13.4个和每条13.2个,并使杯状细胞数量增加至接近正常组水平(每条95.5个),同时上调紧密连接蛋白-1/3(Claudin-1/3,CLDN1/3)、闭合小带蛋白-1(Zonula Occludens-1,ZO-1)和粘蛋白5.1(Mucin5.1,MUC5.1)基因表达,表达量均提高3~4倍。此外还使Toll样受体4(Toll-Like Receptor 4, TLR4)和核因子κB(Nuclear Factor-κB,NF-κB)基因表达量分别降低47.33%和57.12%,并通过调控炎症因子蛋白表达水平,改善机体免疫稳态。综上,OPN和2’-FL组合应用对改善肠炎斑马鱼肠屏障功能具有显著协同作用,这为婴幼儿配方食品的设计优化提供了理论基础。

    Abstract:

    Osteopontin (OPN) and 2,-fucosyllactose (2,-FL) in human milk are important bioactive components that regulate intestinal immunity and barrier function in infants. To investigate the synergistic effects and mechanisms of OPN and 2,-FL combinations on intestinal barrier function, a soybean meal-induced zebrafish larval enteritis model was established. The study intervened with OPN and 2,-FL monomers and combinations at different ratios (1:18~1:1, m/m) and measured the number of intestinal neutrophils and goblet cells, expression levels of inflammation- and barrier-related genes, and protein expression levels of key cytokines. The results demonstrated that 1:9~1:3 (m/m) OPN:2,-FL ratio significantly reduced the number of neutrophils to 13.4 and 13.2 per larva and increased the number of goblet cells to a level close to that in the normal group (95.5 per larva). Meanwhile, the gene expression levels of claudin-1/3, zonula occludens-1, and mucin 5.1 were upregulated by 3~4 fold. Additionally, the gene expression levels of toll-like receptor 4 and nuclear factor-κB were reduced by 47.33% and 57.12%, respectively. The immune homeostasis of the organism was improved by regulating the inflammatory cytokine protein expression levels. In summary, the combination of OPN and 2,-FL significantly synergistically improve intestinal barrier function in zebrafish larvae with enteritis, providing a theoretical basis for the optimization of infant formula design.
