    为了研究油棕仁油 -2 单不饱和脂肪酸(OPO)对小鼠肠道各段短链脂肪酸(SCFAs)水平的影响及其作用机制,C57BL/6J雄性小鼠被随机分为对照组、低剂量OPO组(LOPO,质量分数2.25%)和高剂量OPO组(HOPO,质量分数9.00%),饲养4周后测定肠道各段SCFAs水平,并采用代谢组学和转录组学分析OPO干预对肠道代谢网络和基因表达的影响。结果表明,OPO干预显著提高了小鼠空肠和结肠中SCFAs含量,特别是乙酸、丙酸和丁酸的质量分数在HOPO干预下分别最高提升至115.59%、273.71%和188.74%;而在回肠,高剂量OPO组乙酸和丁酸质量分数显著降低至39.26%和44.79%。代谢组学和转录组学分析揭示了OPO干预显著影响了脂质代谢、消化吸收和信号转导等通路,特别是“脂肪消化吸收”通路高度富集。该研究表明OPO干预通过优化肠道消化吸收过程,剂量依赖性地提高小鼠肠道不同部位(尤其是空肠和结肠)的SCFAs 水平,为促进宿主代谢健康的干预策略提供了理论依据。

    Abstract:

    The effects of palm kernel oil-2 monounsaturated fatty acid (OPO) on short-chain fatty acid (SCFA) levels in different segments of mouse intestine and its underlying mechanisms were investigated. Male C57BL/6J mice were randomly divided into control, low-dose OPO (LOPO, mass fraction 2.25%), and high-dose OPO (HOPO, mass fraction 9.00%) groups. After 4 weeks of feeding, SCFA levels in different intestinal segments were measured, and metabolomics and transcriptomics analyses were conducted to evaluate the effects of OPO intervention on intestinal metabolic networks and gene expression. The results showed that OPO intervention significantly increased SCFA levels, particularly acetate, propionate, and butyrate, and mass fractions in the jejunum and colon, up to 115.59%, 273.71%, and 188.74%, respectively, under HOPO intervention. However, in the ileum, the acetate and butyrate mass fractions in the high-dose OPO group were significantly reduced to 39.26% and 44.79%, respectively. Metabolomic and transcriptomic analyses revealed that OPO intervention significantly affected pathways related to lipid metabolism, digestion and absorption, and signal transduction, with the "fat digestion and absorption" pathway being highly enriched. This study demonstrated that OPO intervention optimizes intestinal digestion and absorption processes, dose-dependently increases SCFA levels in different mouse intestinal regions (particularly the jejunum and colon), thereby providing a theoretical basis for intervention strategies promoting host metabolic health.
