    为探究青稞酒糟中酚类物质的组成和含量,研究其生物活性,该研究以青稞和不同发酵周期的青稞酒糟为原料,提取酚类物质,测定总酚含量并分析物质构成,比较不同样品之间抗氧化和抑菌活性的差异。结果表明,青稞酒糟中总酚含量为4.72~11.25 mg•g-1,从酒糟提取液中共检测出22种酚类化合物,主要成分包括:阿魏酸、儿茶素、没食子酸、丁香酸、芦丁、对香豆酸和绿原酸等。不同发酵周期的青稞酒糟中,丢糟的总酚含量最高,达11.25 mg•g-1,经检测出的酚类物质总量为1 470.64 μg•g-1,其游离酚和结合酚提取液对DPPH、ABTS+自由基清除能力和FRAP还原能力分别为2 052.72、2 319.20、5 050.71 μg TE•g-1和1 996.89、1 710.69、3 344.82 μg TE•g-1;对黑曲霉和青霉的抑菌圈直径分别为15.55、36.56 mm和13.07、37.78 mm,最小抑菌浓度和最小杀菌浓度均为1.56和3.13 mg•mL-1,具有较强的抗氧化活性和抑菌活性。因此,丢糟中的酚类物质组成和含量丰富,抗氧化和抑菌活性突出,有望在天然植物源保鲜剂和功能性产品中进一步开发应用。

    Abstract:

    To investigate the phenolic composition, content, and biological activities of highland barley fermentation spent (HBFS), phenolic compounds were extracted from highland barley and HBFS obtained at different fermentation stages. Total phenolic content was determined, individual phenolic compounds were identified, and differences in antioxidant and antifungal activities among samples were evaluated. Results showed that the total phenolic content of HBFS ranged from 4.72 to 11.25 mg•g-1. A total of 22 phenolic compounds were identified, with ferulic acid, catechin, gallic acid, syringic acid, rutin, p-coumaric acid, and chlorogenic acid as the predominant constituents. Discarded HBFS exhibited the highest total phenolic content (11.25 mg•g-1) and the highest total detected phenolic concentration (1 470.64 μg•g-1). Both free and bound phenolic extracts from discarded HBFS demonstrated strong antioxidant and antifungal activities. The DPPH and ABTS+ radical scavenging capacities and FRAP reducing power of free phenolics reached 2 052.72, 2 319.20, and 5 050.71 μg TE•g-1, respectively, while those of bound phenolics were 1 996.89, 1 710.69, and 3 344.82 μg TE•g-1, respectively. Antifungal assays showed inhibition zone diameters of 15.55 and 13.07 mm against Aspergillus niger and 36.56 and 37.78 mm against Penicillium spp. for free and bound phenolics, respectively. The minimum inhibitory concentration and minimum fungicidal concentration for both extracts were 1.56 and 3.13 mg•mL-1, respectively. These findings demonstrate that discarded HBFS is a rich source of phenolic compounds with pronounced antioxidant and antifungal activities, indicating its strong potential for development as a natural preservative or functional ingredient in food and pharmaceutical applications.
