    为了探究吲哚-3-丙酸(IPA)对高脂饮食诱导的肥胖小鼠肠道菌群及代谢指标的调节作用,将雄性C57BL/6J小鼠随机分为低脂饮食组(LFD)、高脂饮食组(HFD)和IPA干预组(HFD+IPA)。通过灌胃给药16周后检测各组小鼠的体质量、肝脏及睾周脂肪质量变化,观察脂肪沉积情况;采用ELISA检测血清甘油三酯(TG)和总胆固醇(TC)水平;用16S rRNA测序分析肠道菌群组成及丰度变化。结果显示,与HFD组相比,IPA干预显著降低肥胖小鼠体质量增加(P<0.05),肝脏和睾周脂肪质量分别降低26.30%和29.00%,肝脏指数降低26.98%;血清TG和TC水平分别下降23.11%和19.53%;肠道菌群中阿克曼菌属(Akkermansia)相对丰度增加2倍。综上所述,IPA可能通过调节阿克曼菌属等有益菌群丰度改善肥胖小鼠的代谢异常,为利用色氨酸代谢物靶向肠道菌群干预肥胖提供了理论依据和潜在应用价值。

    Abstract:

    The effects of indole-3-propionic acid (IPA) on gut microbiota composition and metabolic parameters in highfat diet-induced obese mice were investigated. Male C57BL/6J mice were randomly divided into three groups: low-fat diet (LFD), highfat diet (HFD), and IPA intervention (HFD+IPA) groups. After 16 weeks of intragastric administration, body weight, liver weight, and peritesticular adipose tissue weight were measured, and lipid deposition was assessed. Serum triglyceride (TG) and total cholesterol (TC) levels were determined using ELISA. Gut microbiota composition and abundance were analyzed using 16S rRNA gene sequencing. The results showed that, compared with the HFD group, IPA intervention significantly reduced body weight gain (P<0.05). Liver weight, peritesticular adipose tissue weight, and liver index decreased by 26.30%, 29.00%, and 26.98%, respectively. Serum TG and TC levels decreased by 23.11% and 19.53%, respectively. Moreover, the relative abundance of Akkermansia in the gut microbiota increased by approximately 2-fold following IPA treatment. These findings suggest that IPA may alleviate metabolic abnormalities in obese mice by modulating the gut microbiota, particularly by increasing beneficial bacteria such as Akkermansia. A theoretical basis and potential application value for targeting gut microbiota with tryptophan metabolites in obesity intervention is provided.
