    为优化仙人草中总黄酮的提取工艺,并评估其降血糖功效。通过单因素和响应面法优化提取条件,确定最佳提取工艺:乙醇体积分数86%,料液比1:18(g/g),超声时间36 min,超声温度66 ℃。在此条件下,理论总黄酮得率为2.15%,实际得率2.17%,相对误差0.93%。4种因素对仙人草总黄酮提取率的影响程度为:超声时间>超声温度>乙醇体积分数>料液比,超声时间与超声温度间的交互作用对总黄酮提取率的存在显著影响(P<0.05)。体外降血糖实验表明,6.0 mg/g仙人草总黄酮对α-葡萄糖苷酶活性的抑制率为72.18%和8.0 mg/g仙人草总黄酮对α-淀粉酶活性的抑制率为64.57%;10 μg/g仙人草黄酮能显著(P<0.05)提高肝脏细胞HepG2的葡萄糖消耗量,并促进肝脏细胞糖原的合成(提高45.4%)。这表明仙人草黄酮具有良好的辅助降血糖功效,该研究结果可为仙人草总黄酮的开发和利用提供理论依据。

    Abstract:

    The extraction process of total flavonoids from Mesona chinensis Benth was optimized, and their hypoglycemic efficacy was evaluated. The optimum extraction conditions were explored using single-factor tests and response surface methodology. The optimum conditions were determined as follows: ethanol concentration, 86%; liquid-solid ratio, 1:18 g/g; ultrasonic extraction time, 36 min; and ultrasonic extraction temperature 66 ℃. Under the above optimum conditions, the predictive extraction rate of total flavonoids from M. chinensis was 2.15%, while the actual extraction rate was 2.17%, with a relative error of 0.93% between the actual and predictive yields. The effects of the four factors on the extraction rate of total flavonoids from M. chinensis were as follows: ultrasonic extraction time>ultrasonic extraction temperature>ethanol concentration>solid-liquid ratio. Additionally, the interaction between ultrasonic extraction time and ultrasonic extraction temperature had a significant (P<0.05) effect on the extraction rate of total flavonoids. The results of in vitro hypoglycemic test showed that the effect of total flavonoids (6.0 mg/g) on the inhibition rate of α-glucosidase activity was 72.18%, whereas the effect of total flavonoids (8.0 mg/g) on the inhibition rate of α-amylase activity was 64.57%. In addition, 10 μg/g of total flavonoids could significantly (P<0.05) increase the glucose consumption of Human Hepatocellular Carcinoma Cell Line cells and promote liver glycogen synthesis (increased by 45.4%). These findings indicated that the total flavonoids from M. chinensis exert a notable auxiliary hypoglycemic effect. The results in this study could provide a theoretical basis for further exploration and potential utilization of the total flavonoids from M. chinensis.
