    该研究采用超声辅助95%乙醇提取法制备了杨树桑黄与粗毛纤孔菌醇提物(SEP与IEP),深入对比了二者在活性成分、多酚组分及体外抗氧化与潜在降血糖活性上的差异,并对其进行了相关性分析。结果显示,SEP中总酚与总甾醇的质量分数分别为33.06%和7.78%,显著高于IEP(P<0.05);而IEP的得率(18.59%)和总黄酮质量分数(25.53%)显著高于SEP(7.96%和13.90%)(P<0.05)。SEP富含原儿茶酸、原儿茶醛和香草酸,含量分别为196.07、142.17和138.77 μg/mg,显著高于IEP(P<0.05),且SEP中7种酚类化合物总含量更高。此外,SEP的DPPH、ABTS+、羟基自由基清除能力及还原力和抗脂质过氧化能力均优于IEP,且SEP对α-葡萄糖苷酶的抑制作用强于IEP(IC50值分别为0.44和0.62 mg/mL);但IEP(IC50值为0.65 mg/mL)对α-淀粉酶的抑制能力比SEP更强(IC50值为0.46 mg/mL)。相关性分析结果表明,酚类化合物与抗氧化活性和酶抑制能力之间相关性较强。综上,尽管SEP的抗氧化活性优于IEP,但二者均具备显著的降血糖潜力,鉴于IEP的高得率,它可能更适宜成本控制下的降血糖产品开发。该试验结果能够推动桑黄资源的综合利用和深度开发。

    Abstract:

    In this study, the ethanolic extracts of S. vaninii (SEP) and I. hispidus (IEP) were prepared by the ultrasonic-assisted extraction method using 95% ethanol, and their active components, major polyphenolic content, in vitro antioxidant activity, and potential hypoglycemic effects were analyzed and compared, with their correlations were analyzed. The results showed that the mass fractions of total phenolics and total sterols in SEP were 33.06% and 7.78%, respectively, which were significantly higher than those in IEP (P<0.05). However, the extraction yield (7.96%) and the mass fraction of total flavonoids (13.90%) of SEP were lower than those of IEP (18.59% and 25.53%, respectively) (P<0.05). Compared with IEP, SEP had higher contents of protocatechuic acid (196.07 μg/mg), protocatechuic aldehyde (142.17 μg/mg), and vanillic acid (138.77 μg/mg) (P<0.05), and a highertotal content of seven phenolic compounds. Moreover, SEP exhibited higher DPPH, ABTS+ and hydroxyl radical scavenging activities, reducing power, anti-lipid peroxidation activity, and stronger inhibitory activity against α-glucosidase (IC50=0.44 mg/mL) compared with IEP (IC50=0.62 mg/mL). However, IEP showed stronger inhibitory activity against α-amylase (IC50=0.46 mg/mL) than SEP (IC50=0.65 mg/mL). The correlation analysis revealed a strong correlation between the phenolic compounds and the antioxidant activities and enzyme inhibitory capacities. In summary, although SEP exhibits higher antioxidant activity and α-glucosidase inhibitory activity than IEP, both exhibit significant hypoglycemic potential. Considering the high yield of IEP, IEP may be more suitable for the development of hypoglycemic products due to its low cost. These results may promote the comprehensive utilization and in-depth development of Sanghuang resource.
