Optimization of High-density Culture and Lyoprotectant for Lactobacillus plantarum BF_15 from Breast-fed Infants

    摘要:

    为推动具有免疫调节、降胆固醇等多种益生特性且遗传背景清晰的婴儿源植物乳杆菌BF_15产业化进程,该研究通过单因素结合正交试验对培养基组分和培养条件进行优化,单因素结合一定复配比对冻干保护剂优化。结果表明:MRS培养基中的活菌数2.30×109 CFU/mL,通过单因素结合正交试验得到最优培养基组分组合:低聚果糖30 g/L、氮源(大豆蛋白胨:酵母膏=3:1)30 g/L、无水乙酸钠6 g/L、硫酸锰75 mg/L,活菌数提高到6.93×109 CFU/mL;在最优培养条件:接种量体积分数5%、培养温度33 ℃、初始pH值7.0、溶氧量0 r/min,活菌数提高到8.92×109 CFU/mL,是优化前的3.88倍。最优冻干保护剂配方:脱脂乳100 g/L、牛血清10 ml/L、蔗糖80 g/L、谷氨酸钠20 g/L、酵母浸粉15 g/L、吐温80 10 ml/L,菌株存活率为90.57%,活菌数达到2.45×1010 CFU/mL。冻干菌粉在4 ℃贮藏56 d,存活率82.35%,活菌数达2.02×1010 CFU/mL;经胃肠液后存活率69.70%,活菌数达1.41×1010 CFU/mL。综上,实现了植物乳杆菌BF_15的高密度培养并获得高活菌制剂,为加快产业化进程提供了坚实理论支持。

    Abstract:

    In order to promote the industrialization of Lactobacillus plantarum BF_15 with multiple probiotic properties (such as immune regulation and cholesterol reduction) from breast-fed infants with clear genetic background. In this study, the medium components and culture conditions were optimized through single-factor combined with orthogonal experiments, and the lyoprotectants were optimized through single-factor experiments combined with specific formulation ratios. The results showed that the viable cell count of BF_15 in MRS Medium was 2.30×109 CFU/mL. The optimal medium composition obtained through singlefactor combined orthogonal experiments was: fructooligosaccharides, 30 g/L; nitrogen source (soy protein peptone: yeast extract=3:1), 30 g/L; sodium acetate anhydrous, 6 g/L; manganese sulfate, 75 mg/L. Under these conditions, the viable cell count of BF_15 reached 6.93×109 CFU/mL. The optimal culture conditions: inoculation volume fraction, 5%; incubation temperature, 33 ℃; initial pH value 7.0; dissolved oxygen level, 0 r/min. Under such conditions, a viable cell count of 8.92×109 CFU/mL was achieved, which was 3.88 times that before optimization. The optimal lyoprotectant formulation was: skim milk, 100 g/L; bovine serum, 10 ml/L; sucrose, 80 g/L; sodium glutamate, 20 g/L; yeast extract powder, 15 g/L; Tween 80, 10 ml/L. Under these conditions, the survival rate of BF_15 was 90.57%, with a viable cell count of 2.45×1010 CFU/mL. After a storage at 4 ℃ for 56 days, the survival rate of the freeze-dried bacterial powder was 82.35%, with a viable cell count of 2.02×1010 CFU/mL. Following the gastrointestinal fluid treatment, the survival rate was 69.70%, with a viable cell count of 1.41×1010 CFU/mL. In summary, the high-density culture of Lactobacillus plantarum BF_15 was realized and highly viable bacteria preparations were obtained, which provides solid theoretical support for accelerating the industrialization process.
